DNA ?'s

First of all i tried to search DNA on here and it say's i have to have a 4 letter word....well it got one! I was going to do some more reading before asking but, that didn't happen so here it goes...
I have helped by fostering some rescue dogs from our local dog pound that were walkers. No papers of course. They get from pups, to old old dogs and everything in between. My question is if one would stumble upon a good hunting dog and wanted to find out to prove it was full walker could one test then get them registered?
Here's another example, I have a beagle she isn't reg, and her mate isn't either, both are to young yet to breed but from what i see i would like to make the cross, could i test both of them and then have the background on them to reg. them or the litter?
I have seen single reg. what is that?
OH to much snow to much time to think.....
